Na2S2O3·5H2O 2.5 g
MgSO4·7H2O 0.5 g
CaCl2·2H2O 0.1 g
NH4Cl 0.1 g
KH2PO4 0.1 g
KCl 0.1 g
Dissolve ingredients in 1 L of distilled water, and autoclave. Cool down under to room temperature. Add following solutions aseptically: Trace element mixture 1 ml, Selenite-tungstate solution 1 ml, Bicarbonate solution 30 ml Adjust pH to 7.0-7.2. Dispense the medium into sterile bottles. Seal the bottles with butyl rubber stoppers leaving headspace filled with air.

Sterilized by autoclaving.

Bicarbonate solution

Distilled water100 ml
Saturated with CO2 gas. Tightly sealed in bottles filled with CO2 gas and then sterilized by autoclaving.

Selenite-tungstate solution
NaOH0.4 g
Na2SeO3·5H2O6 mg
Na2WO4·2H2O8 mg
Distilled water1 L
Sterilized by autoclaving.
