"液體培養 將上述成分溶於H2O中再另外加入1 ml CaCl2 (20 mg/mL)及2 ml FeCl3 (25 mg/mL) 接著以氨水調pH至7.2左右最後補水到1L 殺菌 使用前加入已經濾膜過濾之0.7mL Glucose (200 mg/mL) 固體培養(雙層培養法) A.下層 取BHS1 buffer加入Agar使最後Agar濃度達1.5% 殺菌之後待涼 倒plate B.上層 取BHS1 buffer加入Agar使最後Agar濃度達1.5% 再加入colloidal chitin使collidal chitin最後濃度達0.4% 殺菌之後待涼 倒第二層plate Collidal chitin配法 200mL濃鹽酸中放入stir bar,在Hood中轉動,緩慢加入20g chitin攪拌30-35分鐘(此時chitin會溶成粘稠狀) 倒入5L水中(水需預冷到5-10℃)(此時會有白色膠狀物跑出來) 離心之後取沉澱物,再用水洗,離心…… 至整個懸浮液pH>3.5,離心取沉澱物 取少數沉澱物測量水份含量,可算出Collidal chitin的濃度"
