BEEF LIVER MEDIUM for Anaerobes

Peptone10.0 g
K2HPO41.0 g
Beef liver (cut in small pieces)500.0 g
Soak beef liver with 1 L tap water overnight in refrigerator. Skim fat off top. Sterilize at 121°C for 10 minutes. Filter through cheesecloth. Save meat. Add peptone and K2HPO4. Adjust pH to 8.0. Filter through filter paper and make up volume to 1.0 L with tap water. Boil broth and cool under 100% N2.Place a small amount of CaCO3 in each tube, add meat to a depth of 1/2 inch, cover with broth to a total depth of 2 inches.
